Depends on what genres you like I suppose. I really like the following:

* Golden Axe

* Earthworm Jim 1 & 2
* Eternal Champions
* Cool Spot
* Gairies
* Arrow Flash
* Forgotten Worlds

Those are just some of my favorites.
I suggest most of those already posted above me plus a few more that aren't listed or not really listed:

- Landstalker or Crusader of Centy (especially if you like a Zelda styled game)

- Shining Force 1 and 2 (really good strategy RPGs, especially the second game)

- Ranger X if you want a cool action game that was on the later years of the Genesis and pushes the console graphically.

- M.U.S.H.A if you like challenging shooters.

- Pirates! Gold if you want something totally original and different then anything else the Genesis has to offer. Similar to the more recent Pirates games by Sid Meier

- Mutant League Football and Hockey for fun and crazy sports games for the Genesis.

- NHL 94- still probably one of the best sports games ever created. Tight controls and just so much fun to play against a friend. Started the one-timers.

- Adventures of Batman and Robin for frantic and exciting 2 player action. Great Soundtrack as well (Jesper Kyd of Assassins Creed series and Hitman series).

- Ghouls N' Ghosts -Perfect Arcade ports to a grueling yet satisfying game.

- Gunstar Heroes - one of the best games of the 16 bit generation as a whole. Feels like Contra on Steroids with anime influence.

- Herzog Zwei or Dune II Battle for Arrakis if you like Strategy games and don't mind micro managing with a controller versus a mouse. (or you could try and track down the mouse for the Genesis but I'm not sure if it would work with these games or not).

- Rocket Knight Adventures or Sparkster - The series just got a reboot on for download on current consoles and it might be worth checking out the history of the series.

- Toe Jam and Earl is a good 2 player game. Wouldn't really recommend the second one (panic on planet funkotron) so much though.

-Mega bomberman- Pretty much like any other Bomberman game, so if you like the series look to grab that one.

- Aladdin - A very good platform/action game if you liked the movie at all in the slightest, this is surprisingly a fantastic game on the Genesis.

-Warsong - An overlooked RPG that is actually Langrisser in Japan. A great addition to your collection to sit up there with Shining Force and Shining Force II. Might be a little harder to find this one now.


Might add more, but there are a ton more to still choose from. :)
